Omega-3 fatty acids, particularly EPA and DHA, demonstrate powerful effects on Treg restoration and stability through multiple mechanisms. These essential fats help reduce ceramide accumulation in tissues, which normally suppress FOXP3 expression - the master transcription factor critical for Treg development and function. By inhibiting the enzymatic activity responsible for generating long-chain ceramides, omega-3s alleviate this suppression and promote FOXP3 restoration, thereby supporting healthy Treg populations [30]. Additionally, omega-3 fatty acids increase fatty acid oxidation, which supports the mitochondrial bioenergetics essential for naïve T cell commitment to Treg production [29][32].

Clinical research demonstrates that omega-3 supplementation can restore Treg function even after systemic inflammation has depleted these crucial immune regulators. In experimental studies, mice fed inflammatory high-fat and high-sugar diets developed systemic inflammation accompanied by significant reductions in Treg populations. However, omega-3 fatty acid supplementation resulted in remarkable restoration of both active and functional Tregs, highlighting their therapeutic potential in reversing immune dysfunction [29][32].

Beyond direct Treg support, omega-3 fatty acids address the underlying inflammatory environment that drives Treg dysfunction.
